CFDA(5(6)-Carboxyfluorescein diacetate,CAS:124387-19-5)常用来检测细菌活性和代谢活性。多与细胞核染料碘化丙啶(PI)或DAPI联合使用,以下列出此探针的一些应用文献,仅供交流学习用。

另外有资料显示,革兰氏阴性菌用CFDA染色所得的荧光强度会低于革兰氏阳性菌,原因在于,革兰氏阴性菌的外膜结构会在一定程度上阻挡CFDA进入细胞。对于革兰氏阴性菌染色,可使用添加含EDTA的染色缓冲液(比如0.1M-Phosphate buffer (pH8.5), 5%(w/v) NaCl, 0.5 mM EDTA)或戊二醛(参考文献:Morono Y et al. 检测背景:CFDA是一种比较流行的、灵敏的荧光底物用于预估活微生物(细菌、酵母或真菌)的酯酶活性。细胞质内的酯酶活性是微生物生理活性的一种指标。通过CFDA-DAPI双染色方法同时确定总细菌数和具有生理活性的细菌数(具有酯酶活性)。

检测对象:500ml透析液样本的细菌细胞

检测方法:Bacterial cells in 500 ml of dialysis fluid samples were vacuum-filtered onto polycarbonate black filters and then incubated in staining buffer [0.1 mol/l phosphate buffer (pH8.5), 5% (w/v) NaCl, 0.5 mmol/l EDTA (ethylenediaminetetraacetic acid)] containing DAPI (final concentration, 1 μg/ml) and CFDA (final concentration, 150 μg/ml) in a filtration funnel for 5 min at room temperature (∼25°C). The staining buffer in the funnel was then removed by vacuum filtration. The filters were mounted on glass slides with immersion oil and fluorescing cells were counted at 1000 × magnification using an E-400 epifluorescence microscope equipped with a 100 W Hg lamp.

CFDA SE细胞增殖与示踪检测试剂盒(CFDA SE Cell Proliferation and Cell Tracking Kit)是一款基于荧光探针CFDA, SE,用于细胞增殖检测和细胞荧光示踪的检测试剂盒。此方法目前广泛替代MTT法或者3H-胸腺嘧啶核苷掺入法(3H-thymidine incorporation)等细胞增殖检测方法。本试剂盒包含粉末形式的CFDA, SE探针,细胞培养级的溶剂,以及细胞标记液,使用方便,操作简单。按照每个样本的标记体积为1ml来算,我司(金畔生物)提供两种规格的试剂盒,分别可检测500个和1000个样品。

CFDA, SE,英文全名5(6)-Carboxyfluorescein diacetate N-succinimidyl ester,CAS NO. 150347-59-4,是一种稳定的、细胞膜渗透性的非荧光染料,由两个醋酸基团和一个琥珀酰亚胺酯(SE)官能团组成的一个荧光分子。一旦主动扩散进入细胞,其醋酸基团被胞内酯酶切割,生成荧光素酯CFSE。CFSE具有高度荧光,且能通过其琥珀酰亚胺酯基团共价结合到细胞内的蛋白氨基基团。正是这个共价偶联反应,CFSE能够极其长期的保留在细胞内,长达数个月。另外,归因于这一稳定连接,一旦染料进入细胞,不会转移到邻近细胞。无活力细胞仍然是非荧光。而活细胞一旦分化,CFSE能够很均匀的分散到子细胞中,每分裂一次子代细胞约能得到亲本细胞1/2的荧光强度,因此,通过流式细胞仪对荧光强度的检测,能够依次分选出未分裂细胞,分裂一次的细胞(1/2荧光强度),分裂两次的细胞(1/4荧光强度),分裂三次的细胞(1/8荧光强度),以及以此类推其他分裂次数的细胞。CFSE可以检测分裂多达8次或更多次数的细胞增殖。

CFSE广泛用于细胞增殖和体内细胞示踪研究。CFSE的最大激发和发射波长分别为492nm和517nm,标记细胞后呈绿色荧光。使用流式细胞仪检测可用FL1检测通道。也能用荧光显微镜观察,使用FITC滤片。

保存与运输方法

保存:-20ºC保存,开封前1年有效,其中组分A(CFDA SE粉末)需严格避光,组分B避免强光照射。

运输:冰袋运输。

注意事项

1) CFDA SE溶剂在低温(如4℃,冰浴)等条件下会凝固而粘在离心管底、管壁或盖子上,可将其置于25℃温育片刻直至全部融解后使用。

2) CFDA SE粉末经溶剂溶剂配制成储存液后一个月内用完最好,最多不超过三个月。因CFDA SE容易被水解,并在水溶液中变质。使用过程中尽量避免接触水。而在标记过程中接触到水是在允许范围内。

3) 荧光染料均存在淬灭问题,操作和储存过程中需注意避光,以减缓荧光淬灭。

使用方法

1. CFDA SE储存液(1000X)制备

a) 将低温保存的CFDA SE粉末置于室温回温至少20min,之后短暂低速离心,让所有粉末都掉落至管底。同时也提前将CFDA SE溶剂置于室温或25℃水浴片刻直至充分融解。

b) 吸取一定量CFDA SE溶剂(比如400μl)到CFDA SE粉末中,充分溶解,短暂低速离心,之后全部转移到剩下的CFDA SE溶剂中,混匀后,此即为CFDA SE储存液(1000X)。此配制过程需要避光。

c) 按照单次用量分装,用铝箔纸包好后,置于≤-20℃避光干燥保存,最好一个月内用完,最长不超过三个月。-70℃保存能适当延长保存周期。

2. CFDA SE标记液(1X)制备

准备适量无菌的细胞培养级去离子水,根据实验需要用量,配制适量的CFDA SE标记液(1X)。比如,取10ml 细胞标记液(10X),加入90ml 去离子水,充分混匀后,即得到CFDA SE标记液(1X),短期置于4℃保存,长期不用可置于-20℃保存。

3. 染色步骤

a) 室温300×g离心细胞5min,吸掉上清。

b) PBS或其他生理缓冲液清洗细胞以去除任何残留的血清蛋白。之后再同上离心吸掉上清。

c) 用1ml CFDA SE标记液(1X)重悬细胞,调整密度为1-5 x 106cells/mL,转移到15ml离心管内。

d) 用CFDA SE标记液(1X)将CFDA SE储存液(1000X)稀释到2X。比如,取2μl CFDA SE储存液(1000X)到1ml CFDA SE标记液(1X),混匀后即为CFDA SE储存液(2X)。

e) 将1ml CFDA SE储存液(2X)加入到步骤c)中含1ml 待标记细胞的15ml离心管内,轻轻混匀。

f) 37℃避光孵育15min。【具体的孵育时间可根据实际情况做适当调整,常用孵育时间10-30min。】

g) 立即加入10ml完全细胞培养液(含血清)来淬灭染色过程,室温颠倒几次混匀。

h) 室温离心去上清,再用5-10ml完全细胞培养液清洗一次。

i) 再加入5-10ml完全细胞培养液,37℃孵育5min,以促进CFDA, SE在细胞内的充分反应和未反应的CFDA, SE重新回到完全细胞培养液中。离心去上清,完成最后一次清洗。

j) 之后用完全细胞液重悬细胞,此时可用荧光显微镜(Ex/Em=492/517nm,FITC滤片)或流式细胞仪(FL1通道)来观察标记效果。或开始用药物刺激处理或继续常规培养。经适当时间后用流式细胞仪检测细胞增殖,或用于特定目的的细胞示踪。标记细胞也可用于活体动物的移植,并用荧光进行示踪。标记的细胞呈绿色荧光。

常见应用和数据示例:

根据荧光信号的稀释来检测细胞增殖和分化(示踪)(Cell proliferation and cell division by Dye Dilution Method),此方法主要用来体内外检测淋巴细胞(T细胞和B细胞)的增殖,也能用来检测NK细胞,成纤维细胞甚至细菌。也有用来研究可移植造血干细胞的增殖。
